IL-17E/IL-25

Interleukin-17E (IL-17E)/IL-25 is a member of the IL-17 family of cytokines that also includes IL-17A, IL-17B, IL-17C, IL-17D, and IL-17F. IL-17E is a proinflammatory cytokine and has approximately 16% sequence similarity with IL-17A. However, unlike IL-17A, which promotes TH1 based inflammation, IL-17E primarily stimulates the TH2 cytokine pathway. In cell cultures, IL-17E activates NF-κB and stimulates the production of the proinflammatory chemokine IL-8. Studies in transgenic mice show that overexpression of murine IL-17E (mIL-17E) up-regulates the gene expression of many TH2 cytokines such as IL-4, IL-10, and IL-13, resulting in multi-organ inflammation. High levels of IL-17E causes other immunological changes, including raised serum levels of IL-13 and IL-15, accompanied by pathological changes in different tissues and epithelial cell hyperplasia. IL-17E plays an important role in immunity against helminth infections. IL-17E is also critical in inducing allergic inflammation by promoting TH2 cytokines.
