Tumor Necrosis Factor-α (TNF-α), also known as tumor necrosis factor ligand superfamily member 2 (Tnfsf2) and cachectin, is a transmembrane protein that oligomerizes intracellularly to form a non-covalent homotrimer. The membrane-bound soluble portion of the homotrimer is cleaved by TACE/ADAM17 to form TNF-α. The homotrimer binds to the receptors, TNF RI and TNF RII, both of which are also expressed as homotrimers. TNF-α is mainly secreted by macrophages and a variety of other cell types under pathogenic conditions. Upon receptor binding, TNF-α has been shown to trigger diverse cell signaling pathways including apoptosis, proliferation, differentiation, chemoattraction, hypothalamic regulation, and cytokine production. TNF-α also stimulates IL-1 and induces cachexia, which causes fever. The intracellular form of TNF-α induces IL-12 production in dendritic cells. TNF-α can induce sepsis and inflammation as well as inhibit tumorigenesis and viral replication.
