Macrophage colony-stimulating factor (M-CSF), also called CSF-1 (colony-stimulating factor 1), is a hematopoietin that regulates the proliferation, survival and differentiation of mononuclear phagocytes, tissue macrophages and osteoclasts. All forms of M-CSF are highly glycosylated, with the secreted forms more glycosylated than the cell surface forms. It is produced by several cell types including fibroblasts, stromal cells, astrocytes, osteoblasts, activated macrophages, B-cells, T-cells and epithelial cells of the endometrium. Certain neoplastic cell lines, such as leukemic, pancreatic and lymphoma cell lines synthesize M-CSF. M-CSF is produced in high levels by uterine epithelial cells during pregnancy. High levels of M-CSF are observed in patients with certain neoplastic and reproductive disorders. Liver disease, hepatic injury, haemophagocytic syndrome, thalassemia and amyloidosis are also associated with increased M-CSF levels. M-CSF is elevated in ischemic heart disease. Circulating M-CSF levels has been suggested as a marker for ovarian, breast and endometrial cancers. M-CSF has also been associated with the pathogenesis of atherosclerosis.
