For the sample incubation step, most MSD assays use between 25-50 μL/well sample volumes which is optimal for the suggested protocol (with 1-2 hour sample incubation). Increasing sample volumes may yield higher signal, however, higher sample volumes may need longer incubation times to reach equilibrium.
