Osteopotegerin (OPG), also known as osteoclast inhibitory factor (OCIF), is a glycoprotein member of the tumor necrosis factor receptor superfamily expressed by osteoblasts. Osteopotegerin plays a major role in inhibiting the differentiation and activation of osteoclasts, essential for bone resorption. Known as a decoy receptor, osteopotegerin inhibits the binding of the receptor activator of nuclear factor kappa B ligand (RANKL) to its receptor RANK and thus preventing the recruitment, proliferation and activation of osteoclasts. Abnormalities in RANKL/RANK/osteopotegerin system can lead to osteoporosis, Paget’s disease, rheumatoid arthritis, and bone loss in cancers.
