The characteristic beta amyloid plaques observed in Alzheimer’s Disease are produced by the processing of amyloid precursor protein (APP) into multiple protein fragments. The extracellular protein, sAPPα is released upon APP cleavage by α-secretase, which precludes formation of the Aβ peptides and amyloid plaques, as the cleavage site lies within the Aβ peptide sequence. sAPPβ is the extracellular protein released upon APP cleavage by β-secretase, and the remaining fragment of APP, C99, is cleaved by γ-secretase to form the Aβ peptides. The Swedish variant of APP is cleaved by β-secretase for form sAPPβ sw, and contains a two amino acid substitution (APP 670-671) which renders it more likely to undergo β-secretase cleavage than the wild type protein and results in increased production of the amylyoidogenic Aβ peptides.
