Sclerostin, the product of the SOST gene, is a secreted glycoprotein produced by osteocytes. Sclerostin is expressed highly in bone, cartilage, kidney, and liver. Sclerostin is an important regulator of bone remodeling and homeostasis. Sclerostin has been shown to bind to Wnt coreceptor LRP5/6 inhibiting Wnt signaling which plays a key role in the regulation of bone formation. Sclerostin may also promote apoptosis of bone cells, further inhibiting bone growth. Mutations of sclerostin are associated with sclerosing bone dysplasia: sclerosteosis and van Buchen disease.
