Interleukin-17C (IL-17C), an autocrine cytokine, is a member of the IL-17 cytokine family. IL-17C influences innate immune responses in the epithelium, including the expression of proinflammatory cytokines and chemokines. IL-17 activity is mediated by binding its receptor, IL-17RE/A, which results in the activation of the NF-κB and MAPK pathways. IL-17C in conjunction with IL-17E has been reported to promote Th17 cell development. Increased IL17C/IL17RE signaling may indicate a higher susceptibility to autoimmune disorders. Studies in chimeric mice demonstrate that IL-17C activates the production of TNF-α, a cytokine closely associated with rheumatoid arthritis (RA). These studies suggest that IL-17C, along with IL-17A and IL-17B, plays a crucial role in the pathogenesis of inflammatory arthritis.
