Poly (ADP-ribose) polymerase (PARP) is a nuclear enzyme activated by DNA damage and was one of the first identified death subsrates for caspase-3 in apoptosis. Active PARP transfers ADP-ribose from NAD+ to nuclear proteins, and this modification has been implicated in many fundamental processes like DNA repair, chromatin stability, cell proliferation, and cell death. PARP becomes inactive following cleavage, however the small cleavage product does retain its DNA-binding capabilities, thereby blocking access to damaged DNA and inhibiting further DNA repair. This kit provides assay-specific components for the quantitative determination of PARP (cleaved) (Asp214) in human whole cell lysates.
